Cancer as a Metabolic Disease

Thomas Seyfried is a professor and cancer researcher whose work has challenged the dominant view of cancer as primarily a genetic disease. Building upon the work of Otto Warburg, he has argued that impaired mitochondrial respiration and altered cellular energy metabolism lie at the origin of malignancy, while many of the genetic mutations observed in tumour cells arise downstream from that metabolic dysfunction.

His framework focuses on the defining energetic characteristics shared across diverse tumour types. Cancer cells reduce their reliance on oxidative phosphorylation and increase their dependence on fermentation pathways. Within this model, glucose and glutamine function together as the principal fermentable fuels sustaining tumour growth and survival.

Seyfried has placed particular emphasis on glutamine as the overlooked component of Warburg's theory. Restricting glucose alone may be insufficient because tumour cells can continue drawing energy from glutamine metabolism. His work therefore argues for coordinated therapeutic strategies that alter the tumour's broader metabolic environment rather than targeting only one pathway.

This metabolic interpretation has significant therapeutic implications. Nutritional ketosis, glucose restriction, glutamine targeting, hyperbaric oxygen therapy, and related interventions can be understood as attempts to place pressure on tumour metabolism while preserving the energy requirements of healthy tissues.

Seyfried developed this approach further through the PRESS-PULSE framework. Sustained metabolic pressure is combined with carefully timed therapeutic interventions intended to exploit the metabolic vulnerabilities of cancer cells.

His work has also encouraged renewed examination of the somatic mutation theory of cancer, mitochondrial transfer experiments, the Warburg effect, and the relationship between chronic inflammation, damaged respiration, and malignant transformation.
